Abstract

(EN) The present invention relates to a method for predicting the risk of having or developing Intracranial aneurysms (IA) in a subject, by identifying at least one mutation in an angiogenic protein, such as Angiopoietin-Like 6 (ANGPTL6). In particular, inventors identified one rare nonsense variant (c.1378A>T) in the last exon of the ANGPTL6 gene which encodes a 10 circulating pro-angiogenic factor mainly secreted from the liver shared by the 4 tested affected members of a large pedigree with multiple IA carriers. They showed a 50% reduction of ANGPTL6 serum concentration in heterozygous c.1378A>T carriers compared to non-carrier relatives, due to the non-secretion of the truncated protein produced by the c.1378A>T transcripts. They observed a higher rate of individuals with a history of high blood pressure 15 among affected versus healthy carriers of ANGPTL6 variants, suggesting that ANGPTL6 could trigger cerebrovascular lesions when combined with other risk factors such as hypertension.

(FR) La présente invention concerne un procédé de prédiction du risque qu'un sujet développe ou présente un anévrisme intracrânien (IA), par identification d'au moins une mutation dans une protéine angiogénique, telle que la protéine 6 de type angiopoïétine (ANGPTL6). En particulier, les inventeurs ont identifié un variant non-sens rare (c.1378A >T) dans le dernier exon du gène ANGPTL6 qui code un facteur proangiogénique circulant principalement sécrété par le foie partagé par les 4 membres affectés testés d'une large généalogie à multiples porteurs de IA. Les inventeurs ont montré une réduction de 50 % de la concentration sérique d'ANGPTL6 dans des porteurs hétérozygotes c.1378A > T comparés à des parents non porteurs, en raison de la non sécrétion de la protéine tronquée produite par les transcrits c.1378A > T. Ils ont observé un taux plus élevé d'individus ayant un historique de pression sanguine élevée parmi des porteurs sains contre des porteurs affectés de variants d'ANGPTL6, suggérant que l'ANGPTL6 pourrait déclencher des lésions cérébrovasculaires lorsqu'elle est associée à d'autres facteurs de risque tels que l'hypertension.
